You may need to an a reference to the correct assembly if you swap to 4.5:

/Library/Frameworks/Xamarin.Mac.framework/Versions/Current/lib/mono/4.5/OpenTK.dll

Does that "fix" this issue?

I have checked this issue again by adding a reference to the correct assembly after  swaping to 4.5:

/Library/Frameworks/Xamarin.Mac.framework/Versions/Current/lib/mono/4.5/OpenTK.dll

But this fix does't work.

An update to this issue:-

We are getting same behavior with following XM Samples also:

1. GlFullScreen
2. MonoMacGameWindow
3. OpenGLLayer
4. OpenGLViewSample
5. OpenGL-Nehe (All Samples)

Sample Info: mac-samples65db48152c5cd289433c3e57faae8f6c4f85fd15

I have checked this issue with the following master build: 
xamarin.mac-2.5.0.2850_7f693655fed61f547446c15e9ba390e74fc65654

and I observed that I am successfully able to build and run all the Xamarin Mac sample that are listed in the comment 3, using Mobile/ .NET 4.5 as target framework.

This issue has been fixed in master build , I will Re-Verify this issue when fix will merged in release branch.

Sample Info: e8e01eff16d850bf071981f4f31829c9f20ff500

I have checked this issue with the latest stable build and observed that this fixed has been merged in release build. Now user is able to build and run the XM GLSLShader sample application using Unsupported/Mobile/ .NET 4.5 as target framework.
